Characterization and Phylogenetic Analysis of a Novel GH43 β-Xylosidase From Neocallimastix californiae

Degradation of lignocellulosic materials to release fermentable mono- and disaccharides is a decisive step toward sustainable bio-based economy, thereby increasing the demand robust highly active lignocellulolytic enzymes. Anaerobic fungi phylum Neocallimastigomycota are potent biomass degraders harboring huge variety such Compared cellulose, hemicellulose degradation has received much less attention; therefore, focus this study been enzymatic xylan anaerobic as these organisms produce some most effective known hydrolytic We report heterologous expression GH43 xylosidase, Xyl43Nc, GH11 endoxylanase, X11Nc, from fungus Neocallimastix californiae in Escherichia coli . The enzymes were identified by screening putative proteome. Xyl43Nc was against 4-Nitrophenol-xylopyranosides with K m 0.72 mM, k cat 29.28 s −1 , temperature optimum 32°C pH 6. When combined, X11Nc released xylose beechwood arabinoxylan wheat. Phylogenetic analysis revealed that shares common ancestry Spirochaetes groups separately Ascomycete sequences our phylogeny, highlighting importance horizontal gene transfer evolution fungi.
